Annexin IV Picobandâ„¢ antibody detects phosphatidylserine (PS) residues exposed on the outer surface of cells during the process of programmed cell death, also known as apoptosis. In live healthy cells, PS residues reside within the inner leaflet of the plasma membrane and are inaccessible to annexin V binding. As apoptosis progresses, the integrity of the membrane destabilizes, and PS residues move to the outside of the plasma membrane where they become accessible to annexin V binding.

This allows annexin V to identify the cell as dead, which is then characterized by a fluorescence signal that is subsequently measured using flow cytometry. Adding a cell impermeant stain, such as propidium iodide (PI) or 7-AAD in conjunction with annexin V conjugates enables apoptosis detection as well as removing false positives resulting from the rupturing of the cell’s plasma membrane.
